The larger dose given at the initiation of treatment in order to quickly achieve the desired blood or tissue final concentration. Amount of drug administered at the onset of treatment to rapidly bring the amount of drug in the body to therapeutic levels.

Mechanical effect of loading, usually cyclic in nature, on the stability of bolted or screwretained joints. Micromotion and fatigue are potential negative occurrences that may result from such loading.

Effects of loading on framework
Load distribution to supporting implants is affected by the relative stiffness of the prosthesis connecting the implants. With a rigid prosthesis, load is transferred relatively equally to all underlying implants. With a more resilient prosthesis, loading tends to be greatest at the implant closest to the point of load application.
